※ NKF3 family introduction

    NKF3 kinases are original defined as new kinase family 3. NFK3 contains two members in human genome, SGK223 and PEAK1. Analysis of PEAK1 shows that PEAK1 contains multiple motifs, domains and consensus phosphorylation sites and PEAK1 localizes to the actin cytoskeleton and focal adhesions. PEAK1 contains kinase activity in vitro and mediate the regulation of cytoskeleton and focal adhesion proteins and undergoes Src kinase-dependent tyrosine phosphorylation in response to cell adhesion or EGF simulation. In addition, PEAK1 expression modulates cell spreading and migration. Furthermore, PEAK1 have been found related to cancers. Studies show PEAK1 can promote anchorage-independent cancer cell growth in vir and tumor progression in mice and PEAK1 expression is up-regulated in human colon cancer and liver metastases. Another family member SGK223 is though to be a kinase and remain to be understood (1).
